Laboratory approach based on GAM-E
The main advantages of GAM-E are simplicity, flexibility, high quality sequence. Based on GAM-E system three especially complex amplicon systems for TCR beta, gamma and delta repertoire profiling have been successfully developed.
GAM-E system is based on three PCRs: PCR-1 captures the target region of interest, PCR-2 adds the elements needed for sequencing (P5, NSP1, UMI and barcode in the left end and P7 in the right end), and PCR-3 does the final generic unbiased amplification based on P5 and P7.
